Interior of the original church (1867-1918)
The Parish of St. Vincent Ferrer was founded in 1867 and is now in its second building on the same site. The first building, once referred to as the "Gothic Barn," was run by the Dominicans, or Order of Preachers, and was built in 1867-1869 to designs by Patrick C. Keely. The adjacent convent, designed by William Schickel and built in 1881, still stands on the corner of 65th Street.

The current building, completed in 1918 and modeled on European Gothic churches, was designed by Bertram Grosvenor Goodhue, one of the leading American church architects of the early 20th century. After the building was completed, a panel of architects tasked with selecting the 50 most beautiful U.S. churches voted St. Vincent Ferrer as the second most beautiful (St. Thomas Church on Fifth Avenue, which won first place, was also one creation of Mr. Goodhue). However, Goodhue considered St. Vincent Ferrer to be his finest work. Exterior limestone carvings by Lee Lawrie and interior fittings, initially incomplete due to lack of funds, were later completed to Goodhue's plans. The building is a federal monument and was also designated by the New York City Landmarks Preservation Commission in 1967.

The organ at St. Vincent Ferrer has evolved from multiple origins over the years. In 1990 James Konzelman rebuilt the gallery organ with three manuals and 30 registers. Then, in 1998, the Schantz organ added 18 ranks (opus 2145) in the choir and provided a new movable four-manual console that controlled the gallery and choir sections. In 2002, Schantz installed a new gallery organ (Opus 2224) with 68 stops and a completely new movable four-manual console in the choir.

One of the more unusual organs in New York City was the five-manual instrument installed at St. Vincent Ferrer in 1926 by the Fratelli Balbiani firm of Milan, Italy. Two consoles were provided: a five-manual moveable console in the choir and an "extra" two-manual console on the choir gallery. The consoles contained 55 flares, an automatic transposer and three pistons for the "automatic pedal combination". Numerous valves and stamps supplemented the five main pistons of the organ. On May 9, 1926, the organ was blessed by Bishop John J. Dunn, followed by a program of music performed by Pietro Yon, honorary organist of the Vatican, and Constantino Yon, organist and choirmaster of the church.

This organ was removed in 1958 and the five-manual console survives in Brooklyn.

The organ in the previous church building was built in 1874 by Hilborne L. Roosevelt of New York City. This was Roosevelt's third three-manual instrument built by the company. Roosevelt used tubular mechanics for the swell and pedal organs, and the pneumatic (Barker) lever was applied to the grand organ and its clutches. The organ was 32 feet wide, 18 feet deep and almost 40 feet high.
